Например, в результате одного из недавних исследований выяснилось, что благодаря этой “мусорной” ДНК возникли млекопитающие, так как эндогенные ретровирусы, или ERV, подстегнули эволюцию плаценты. Это значит, что без “вирусов-зомби” никогда не появились бы люди. Теперь же исследование показало, что эти вирусы могут иметь отношения к онкологическим заболеваниям.

Как известно, злокачественные клетки присутствуют в организме всегда, однако иммунная система их подавляет, поэтому они не развиваются в рак. Онкология же возникает только когда в организме происходят определенные изменения, которые способствуют заболеванию. В частности, известно, что раковые клетки экспрессируют (активируют) множество генов, которые не должны быть включены.
Ученые долгое время не могли понять, что именно “включает” гены, которые помогают развиваться раку. Теперь же исследователи обнаружили, что многие “активаторы” генов происходят от вирусов-зомби.
Как древние вирусы помогают развиваться раку
В ходе исследования ученые проанализировали наборы данных более 20 различных видов рака, и обнаружили, что при этих заболеваниях в активном состоянии находится семейство ERV, называемое длинным терминальным повтором 10 (LTR10). Известно, что этим вирусом наши предки приматы заразились примерно 30 миллионов лет назад.

Обнаружив активность вируса, исследователи детально изучили процессы, которые происходят на генном уровня при колоректальном раке. Как выяснилось, LTR10 регулирует экспрессию генов, которые играют важную роль в возникновении опухолей.
Когда исследователи отключили у мышей гены LTR10 в клетках колоректальной опухоли, вместе с ними были деактивировлись гены, которые вызывают рост рака и делают его устойчивым к терапии. В результате, после отключения LTR10, опухоли мышей стали уменьшаться и поддаваться лечению. Об этом исследователи сообщили в журнале Science Advances.
Почему с возрастом увеличивается риск рака
Напомним, что ученые еще несколько лет назад пришли к выводу, что древние вирусы имеют отношение к раку, однако не знали какое именно влияние они оказывают на опухоль. Теперь же удалось выяснить, что раковые клетки используют вирусы-зомби для изменения паттернов экспрессии генов, и это позволяет опухолям развиваться и прогрессировать. Как показало исследование, только одно семейство ретровирусов способно регулирует до 70 генов, связанных с раком. Сколько таких ретровирусов содержит ДНК человека, ученые сказать не могут.
