Причины возникновения рака
Мутации в ДНК, который приводят к возникновению рака, возникают по разным причинам. Например, это могут способствовать внешние мутагены — продукты с вредными пищевыми добавками, например, мясные деликатесы, табачный дым, загрязненный воздух и т.д. Также мутации вызывают молекулы окислители, которые являются побочным продуктом внутриклеточных процессов. Иногда ошибки в ДНК возникают сами по себе в процессе деления клетки.
Надо сказать, что мутации, связанные с раком, могут возникать в разных участках генома, как кодирующих белках и служебных РНК, так и не некодирующих. По своему действию они тоже бывают разными — одни способствуют возникновение опухоли, а другие оказывают на нее влияние уже после того, как она сформировалась. Но, в любом случае, в результате мутаций клетка перестает выполнять свою функцию и начинает безудержно делиться.
Но как именно эти мутации заставляют здоровую клетку превратиться в злокачественную? Ответ довольно прост — они меняют активность генов. Отсюда можно предположить, что существуют и другие механизмы, которые меняют активность генов без каких-либо изменений последовательности в ДНК.

Как эпигенетика влияет на активность генов
Давно известно, что на активность генов может влиять эпигеном — он способен как простимулировать работу того или иного гена, так и подавить его, не затрагивая при этом генетическую последовательность. Причем эпигенетические изменения могут оставаться с человеком в течение всей жизни.
Эпигеном модифицирует в клетке белки-гистоны, которые влияют на доступность или недоступность некоторых участков генома для других белков, считывающих генетическую информацию. Проще говоря, они могут скрывать часть генома от считывающих белков. Иногда гистоны “упаковывают” некоторые участки генома в “генетический архив”, скрывая его в течение практически всей жизни клетки.
Эпигенетическая модификация, то есть вариация количества и структуры гистоновых белков, могут либо способствовать, либо препятствовать транскрипции тех или иных генов. Эпигенетическая модификация гистонов происходит за счет химических метильных меток, которые к ним прикрепляются. Однако прикрепление меток осуществляется не само по себе, а при помощи других белков Polycomb’ов. То есть без них гистоны не могут получить метильные метки. Но только метилированные гистоны могут упаковывать тот или иной участок ДНК.

Почему рак возникает без мутации
Так как белки polycomb одинаковые у разных организмов, например, млекопитающих и насекомых, ученые провели эксперимент с личинками дрозофил. В ходе исследования они понизили количество некоторых polycomb-белков. Результатом этого стал рост опухолей. То есть клетки личинок потеряли свою специализацию и начали слишком быстро делиться.
В момент, когда этого белка в клетках было мало, ученые заметили, что активность некоторых генов повышалась, что не удивительно. Но самое интересное, что после того, как уровень белка polycomb в клетках нормализовался, опухоли не проходили. При этом ученые заметили, что после нормализации уровня белка, активность генов снова понизилась, но не всех. Некоторые продолжали оставаться активными.
В результате дальнейшего исследования ученые обнаружили цепочку молекулярных сигналов, которые привели к постоянной активности некоторых белков. Эти белки необходимы стволовым клеткам, так как поддерживают их в стволовом состоянии. Кроме того, они стимулируют регенерацию тканей и настраивают определенным образом обмен веществ.

Когда “стволовые” белки все время остаются активными, происходит злокачественное перерождение клеток. Таким образом, эпигенетическая аномалия, связанная даже с временным уменьшением белков-регуляторов polycomb, переводит “стволовые” белки в режим самоподдерживающейся стимуляции.
