Это приводит к параличу мускулатурной системы, в том числе и отвечающей за дыхание. Поэтому, если человеку вовремя не оказана медицинская помощь, он может умереть от остановки дыхания. Но даже после ввода противоядия укус кобры часто вызывает кровоизлияния, что приводит к осложнениям и иногда к смерти. В то же время предыдущие исследования показали, что яд змеи не разрушает клетки сосудов, поэтому причина кровоизлияний оставалась загадкой. Поэтому существующие противоядия не всегда эффективны. Почему противоядие против яда кобры не всегда эффективно
Змеиные укусы по сей день остаются серьезной проблемой здравоохранения, особенно в субтропических регионах. По статистике ВОЗ, ежегодно от укусов змей страдают от 1,8 до 2,7 миллиона человек. При этом от 81 000 до 138 000 случаев оканчиваются летальным исходом. Также часто змеиные укусы приводят к тяжелым последствиям — слепоте, хроническим заболеваниям органов и т.д. Одной из причин летальных исходов и осложнений, как уже было сказано выше, являются кровоизлияния.
В ходе предыдущих исследований воздействия яда на кровеносную систему, ученые использовали двухмерные модели эндотелиальных клеток, то есть клеток, которые выстилают сосуды. Как оказалось, химически они устойчивы к действию яда, поэтому, как уже было сказано выше, причина кровоизлияний оставалась загадкой.

Органоиды и орган на чипе — что это такое
В недавнем исследовании ученые использовали новую технологию, которая называется “орган на чипе”. По сути, они использовали органоид кровеносных сосудов человека совместно со специальным устройством (чипом). Органоид включал в себя эндотелиальные клетки и внеклеточный матрикс, известный еще как физическая структура, которая поддерживает клетки.
Ранее мы уже рассказывали, что органоиды представляют собой трехмерные скопления клеток того или иного типа, выращенные в лабораторных условиях. Несмотря на то, что они представляют собой не полноценный орган человека, значительно расширяют возможности ученых в плане новых исследований. Например, благодаря органоидам ученые могут выяснить как развиваются те или иные органы эмбрионов, тестируют на них противоядия и лекарственные препараты, выясняют как на организм действуют вирусы и т.д.

«Орган на чипе», а точнее сам «чип» создан специально для выращивания различных клеточных культур. Он состоит из микроконтейнеров и микроканалов, что позволяет не только выращивать различные клеточные культуры, но и симулировать работу отдельных систем органов, органа целиком или даже нескольких органов одновременно. Таким образом исследователи смогли создать свою крошечную модель кровеносных сосудов.
Почему яд кобры убивает человека
Как сообщается в исследовании, опубликованном в журнале Scientific Reports, ученые подвергли модель кровеносных сосудов воздействию яда четырех видов змей: индийской кобры, западноафриканской ковровой гадюки, многополосных крайтов и мозамбикской плюющейся кобры. Все они относятся к наиболее ядовитым семействам змей — гадюкам и элапидам.
Также ученые использовали специальные методы визуализации, которые позволяли наблюдать в реальном времени за процессами, происходящими внутри чипа. Как выяснилось, существует два различных механизма воздействия ядов на кровеносный сосуд. В одном случае они вызывают отслоение эндотелиальных клеток от его матрикса, то есть фактически “выбивают” клетки, что вызывает коллапс сосудов. Во втором случае приводят к разрушению мембраны эндотелиальных клеток.
